Hydroxy-ethyl-tetrahydro-(1,4)-oxazine derivs. - whose quaternary salts are antispasmodics for treatment of vascular, digestive and bronchial smooth muscle spasmodic syndromes

2-(2-Hydroxyethyl)-tetrahydro-1,4-oxazines of formula (III) and their quat. ammonium derivs., with R1X (IV) are novel cpds. (where Ar, Ar1 independently = opt. halo substd. aromatic residues, R, R1 independently = 1-4C alkyl and X = an anion of the type Cl-, Br-, I- or CH3SO4-). (IV) are spasmolytics esp. useful for treatment of spasmodic syndromes of smooth muscle or vascular, digestive and bronchial origin, anticholinergics and musculostropics and (III) are intermediates in their preparation. They are administered by usual routes in daily doses of 10-500 mg. A typical prod. is 2-(2,2-diphenyl-2-hydroxyethyl)-4-methyl-tetrahydro-1,4-oxazine prepd. from 2-cyanomethyl-4-methyl-tetrahydrooxazine via the 2-carbethoxymethyl deriv. and reaction with phenylmagnesium bromide. The product may be reacted with methyl bromide to form the quaternary salt.
