Measuring factor XIII content of blood - by determn. of coagulation time after incubation with plasmin and thrombin

Method for determining the fibrin-stabilising factor (factor XIII, I), esp. in human blood, comprises measuring the coagulation time of a sample of the blood. The measurement is pref. carried out in a soln. of a plasma lysate obtd. by incubation with thrombin and plasmin. Then addn. of thrombin distorts the coagulation time and the (I) concn. is determined by measuring the extent of this distortion. A reduced (I) content is esp. revealed by measuring the accelerated coagulation time relative to the normal time. A test kit for the assay comprises a factor XIII-reagent and thrombin in separate containers. Compared with known (e.g. antibody inhibition) procedures, this method is qick, simple and inexpensive, so suitable for routine use. It is useful in diagnosis of congenital (I) deficiency, wound-healing abnormalities and malignant blood disease.
