The biological specimen is illuminated by light from a laser source which may be coherent, partially coherent or incoherent. The output light from the specimen is then examined and split into several light paths to facilitate measurement. The prepared specimen is placed between a condenser lens and an objective lens and illuminated. The wavelength of the light may be altered to improve the contrast of certain subjects. The output light is split into three separate paths by beam splitters. Fourier zones from the objective lens are filtered and reappear in each of the three light paths. Further lenses have a focus point which is the input of a computer.
