Novel process for concn. and purification of Factor VIII (antihaemophilic factor) is claimed. Method comprises (a) extg. a cryopptn. prod. in an aqs. medium of low ionic concn. at approx. ambient temp.; (b) pptg. the fibrinogen from this soln. by a single cooling step. to ca. 1-2 degrees C for =1/2 hr.; (c) sepg. the supernatant liq. contg. =80% of the Factor VIII present in the cryopptd. starting prod; and (d) treating the liq. to ensure long-term preservation of a concentrate of the factor. Factor VIII has therapeutic uses and in the prefd. process is obtd. in high purity with reduced losses by selective pptn. of fibrinogen and its denatured and degraded p prods. Process is shorter than prior art (8hrs cf. 16-24hrs) Pref. a supplementary stage is used comprising adding 3-6% of a polyol to the supernatant liq. of (c) and cooling to ca 0-2 degrees C for 1-2 hrs. to ppte. a further amt. of fibrinogen and its denatured and degraded prods.
