A lyophilised prod. is prepd. in a container which has an external bead around its neck. A closure for the neck includes a top wall from which depends a stopper portion. The stopper portion comprises an upper cylindrical section having a larger dia. than the neck of the container, and a bottom cylindrical section having a smaller dia. The two cylindrical sections are interconnected by a bevelled section. Ports are provided for venting the container during the lyophilisation process. The closure is inserted into the neck of the container in a first position during the lyophilisation process at which the ports are positioned above the neck of the container. Subsequently the closure is inserted to a second position in which a tear-away skir portion extending downwardly from the top wall engages with the bead, and the ports are within the neck to prevent venting. Used in the prepn. of substances such as biologicals, serums, hormones, blood coagulation reagents, and chemistry control reagents which have to be dehydrated by lyophilisation as they are damaged at normal drying temps. The closure allows venting during lyophilisation and prevents tampering after sealing. Also the closure may be removed easily without risk of injury to a user.
