New procedure for measuring the concentration of a substance in a solution comprises (a) contacting a specimen of the solution with an enzyme specific for the substance to be measured and with an appropriate coenzyme, (b) contacting at least the reduced form of the coenzyme resulting from the ensuing reaction with a second or auxiliary enzume, and (c) measuring the amount of molecular oxygen and/or hydrogen peroxide. Used for determination of substances such as glucose in aq. solutions. Enzymatic oxidation of the reduced coenzyme to its original oxidised form causes O2 or H2O2 to be released in an amt. proportional to the amt. of substrate present in the original soln. The amount of O2 or H2O2 can be determined by e.g. fluorescence methods.
