Medium to detect Proteus microorganisms in urine - comprises nutrients, urea, buffer to avoid detecting klebsiella, antibiotics and colour-changing indicator
Medium for a Proteus detecting broth comprises (a) a source of nutrients, (b) urea, (c) buffer to prevent false positive results due to Klebsiella organisms, (d) antibiotics to inhibit the growth of Klebsiella and Staphylococcus aureus and organisms positive to urease and (e) a biological pH indicator reacting to an increase in pH towards alkaline values by a change of colour. Pref. colistin at 4850-5150mg/l is used to inhibit growth of Klebsiella and erythromycin at 4850-5150mg/l to inhibit Staphylococcus aureus. The pref. source of nutrients is Protease-Peptone No. 3 (RTM) used in concns of 0.97-1.03g/l. The medium is used for detection of Proteus in urine as an indicator of urinary tract infection, whilst avoiding false results due to Klebsiella, Staph. aureus etc. The tests using this method are complete in 12-18 hrs. whilst known methods require 36-48 hrs.
