Agglutination type immunological determn. of immunoglobulin antibodies - by adsorbing an anti-human IG on a solid, incubating with the sample and treating the adsorbed fraction with insoluble antigen
Immunological determn. for detecting an immunoglobin antibody, which is specific to an antigen, in body fluid samples comprises (a) adsorbing an immunoglobulin antibody on a solid surface; (b) applying the body fluid sample to the adsorbed immunoglobulin antibody and incubating the mixt; (c) sepg. the non-adsorbed fraction from the adsorbed fraction after incubation is complete (d) treating the adsorbed fraction with an insol. form of the antigen and (e) determining the degree of binding of the antigen to the adsorbed antibody fraction and observing the distribution of the antigen on the surface. Agglutination-type immunological determn. for detecting of immunoglobulin pref. (IgM) antibodies in body fluid, esp. for detection of IgM antibodies associated with infectious toxoplasmosis e.g. acute forms of Toxoplasma gondii infections. The present determn. combines aspects of the direct agglutination (DA) method and the IgM-ELISA (enzyme linked immunosorbent assay) and is more specific and sensitive than the DA method and simpler and easier to operate than the IgM-ELISA determn. The process is partic. useful for serological diagnosis of congenital (neonatal and perinatal) and post-natal acute forms of toxoplasmosis. The anti-toxoplasmic antibodies are present in all body fluids, but blood serum is preferred for post-natal determinations and amniotic or umbilical cord blood serum for congenital diagnosis. The test may be applied to detection of antibodies specific to other organisms, such as heptatitis A virus, German measles, poliomyelitis, Herpes-simplex, cytomegalo-virus, Epstein-Barr virus and various fungi, protozoa and helminths.
