MEASUREMENT OF ACTIVITY OF SERUM ALKALI PHOSPHATASE

PURPOSE:To measure activity of serum alkali phosphatase acurately with high reliability, by using a diethanolamine-hydrochloric acid buffer solution containing Thymolphthalein monophosphoric acid as a substrate. CONSTITUTION:A diethanolamine-hydrochloric acid buffer solution at 9.75- 9.85pH (at 37 deg.C) containing 0.15-0.2wt% Thymolphthalein monophosphoric acid, preferably its sodium salt, or magnesium salt as a substrate is added to a specimen, Thymolphthalein liberated by a conventional precedure is colored under alkali conditions and its absorbance is measured to determine alkali phosphatase activity.
