ENZYMATIC METHOD AND STABILIZED SOLUTIONS FOR DETERMINING TOTAL CHOLESTEROL IN HUMAN SERUM

Abstract of the DisclosureStabilized enzymes useful in the diagnostic assay oftotal cholesterol are prepared by dissolving a salt ofcholic acid in a buffer solution providing a pH within therange of a?out 4 to about 9, to the solution is added acholesterol esterase. The solution is then mixed with apolyhydroxy organic compound and TRITON-X-100. A cholesteroloxidase and a peroxidase are each dissolved in separateportions of buffer solution and introduced into the bufferedsolution containing the cholesterol esterase. 4-aminoanti-pyrine is then added to the solution. The resultant solutionis a stabilized enzyme solution which can be used in thetotal cholesterol assay of a serum sample. The stabilizedsolution can be used in combination with a chromogen diluentsolution which is made by dissolving pnenol and TRITON-X-100in water or a buffer solution. The combination of thestabilized enzyme solution and the chromogen diluent solu-tion provides a solution which has utility in the spectro-photometric assay of total cholesterol.
