PURPOSE:To determine the saccharinity of grain, rapidly, easily and objectively, by saccharifying the starch in the grain with saccharase, and measuring the sugar concentration (%) of the saccharified liquid with a refractometer. CONSTITUTION:Juice obtained by squeezing grains such as corn, etc. is put into a small test tube made of a plastic, added with a saccharase such as alpha-amylase, mixed thoroughly under shaking, and left to stand for 10-15min at room temperature to effect the saccharification of starch. The saccharified liquid is applied to a saccharimeter such as hand Brix refractometer, and the sugar content (%) is measured.
