4-Guanidinomethyl- cyclohexane carboxylates of formula (I) and their salts are new (where R1 is indanyl, indolyl, quinolyl, chromonyl, R2-substd. naphthyl or R3, R4, R5-substd. phenyl; R2 is halo or nitro; R3 is halo, alkyl, nitro, cyano, acetamido, sulphamoyl, benzoyl, phenoxy or trifluoromethyl and R4, R5 independently are H, halo or alkyl). (I) inhibit serine proteases and are anti-allergics. Serine proteases include trypsin, chymotrypsin, thrombin, plasmin, kallikrein and urokinase. (I) inhibit the liberation of histamine, prevent anaphylactic shock and experimental asthma in guinea pigs. (I) have low toxicity (oral LD50 of (I)/R1 is 4-tert-butylphenyl as the hydrochloride of the trans isomer) is 5400-73000 mg/kg in mice and 10800-11000 mg/kg in rats) and are administered parenterally or orally (daily doses of 30-900 mg).
