PURPOSE:To determine beta-hydroxybutyric acid easily and rapidly, by reacting beta-hydroxybutyric acid with NAD in the presence of a dehydrogenase, reducing a tetrazolium salt with the produced NADH, and determining the amount of the produced formazan by colorimetry. CONSTITUTION:beta-Hydroxybutyric acid in a specimen is oxidized to acetoacetic acid with beta-hydroxybutyric acid dehydrogenase in the presence of nicotinamide adenine dinucleotide (NAD). The tetrazolium salt added to the system {e.g. 3,3'- (3, 3'-dimethoxy-4, 4'-biphenylene)-bis[2-( p-nitrophenyl )-5-phenyl-2H-tetrazolium]} is reduced by the reducing NAD (NADH) produced by the above oxidation reaction, by the aid of an electron transfer material (e.g. methyl-1-methoxy-5-methylphenazium sulfate), and the produced colored formazan is determined by colorimetry to determine the beta-hydroxybutyric acid (cf. reaction formula).
