Droplet counter-current chromatography is utilized to isolate and purify 12-O-tetradecanoylphorbol-13-acetate (TPA), phorbol, and the diastereoisomer 4&agr;-phorbol from croton oil. The stationary and mobile phases for the procedure are derived from an equilibrated mixture of hexane-diethylether-n-propanol-ethanol-water, the separated upper layer of which comprises the stationary phase and the bottom layer of which the mobile phase. The TPA and the phorbol isomers are obtained in exceptionally high purity by further treatment with liquid column chromatography and thin-layer chromatography.
