METHOD FOR SEPARATING AND PURIFYING HUMAN LACTOFERRIN FROM HUMAN MILK

PURPOSE:To separate and purify human lactoferrin in high yield and purity, from human milk, by using a specific affinity column. CONSTITUTION:A monoclonal anti-human lactoferrin antibody produced by a hydridoma is immobilized to an insoluble carrier [e.g. Affigel-10(Biolad Corp.), etc.], and is used as an affinity column to separate and purify human lactoferrin from human milk. The above antibody can be prepared by fusing the spleen cell of mouse immunized with human lactoferrin and a myeloma cell, administering the resultant hybridoma to mouse, and recovering and purifying the ascites of the mouse. EFFECT:The objective compound can be separated easily from the other milk components only by passing through a column once, without using pretreatment. USE:Preventive and remedy for the infective diseases of infants caused by iron- demanding pathogen.
