PRODUCTION OF STOCK STRAIN FOR LIVE VACCINE FOR BORDETELLA BRONCHISEPTICA INFECTION AND LIVE VACCINE

PURPOSE:To obtain the titled stock strain having high immunogenicity and suitable for the production of a vaccine having excellent safety, by applying mutagenic treatment to a Bordetella bronchiseptica strain. CONSTITUTION:A Bordetella bronchiseptica strain is subjected to mutagenic treatment [e.g. a combination of high-temperature treatment (42 deg.C) and nalidixic acid treatment] to separate a mutant at least containing a gene label of temperature sensitivity (grows at 42 deg.C), nalidixic acid resistance and negative to thermolabile toxin productivity. The strain is cultured in a Bordet-Gengou agar medium containing nalidixic acid at 42 deg.C and a strain resembling an I- phase strain is fished. The culture in a Bordet-Gengou agar medium containing nalidixic acid and the fishing of I-phase strain are repeated to obtain an I-phase strain having complete capsule antigen and negative to thermolabile toxin productivity and obtain the stock strain for the production of live vaccine for Bordetella bronchiseptica infection.
