Process for the enzymatic hydrolysis of a lignocellulosic substrate in which the hydrolysis is carried out in a series of successive operations, characterised by the fact that the enzyme source used in each operation consists, at least in part, of at least a portion of the solid residue from the preceding operation. A fresh lignocellulosic substrate 6a is supplemented with the residue 9a from a previous enzymatic hydrolysis and the resulting mixture is subjected to an enzymatic hydrolysis in a container 15. The effluent is separated into a sweet juice 5b and a solid residue 4b which is transferred to a subsequent enzymatic hydrolysis operation. Application to the production of sweet juices IMAGE
