A reagent for the detm. of auto-antibodies of microsomatic thyroid glands is prepd. as follows. Tyroid glands removed from patients suffering from goitre are washed with 0.15% saline soln., homogenised, centrifuged for 30 mins. at 1100 x G, then for 10 mins at 10000 G and 9 mins at 78000 G. The sediment is washed and taken up in saline phosphate buffer. Then the suspn. is treated ultrasonically at 10 kHz frequency five times for appx. 1 min. each. The treated suspn. is centrifuged at 3000 x G for 15 mins. and at a volume of antigen of 0.05 g protein/ 100 mls a volume of glutaric acid and tannatee stabilised 4% erythrocyte suspn. is added. The suspn is allowed to stand for 2 hours at 55 deg.C, is washed with a saline phosphate buffer soln. and centrifuged. Finally, a 1% suspn. in saline buffer is prepd. The buffer also contains 1% normal rabbit serum.
