RECOMBINANT HUMAN TISSUE TYPE PLASMINOGEN ACTIVATOR

PURPOSE:To obtain a recombinant human tissue type plasminogen activator (human t-PA) having biological characteristics different from human t-PA existing in the natural world, containing alpha (1-3) bond galactose at the nonreducing end of conjugate type saccharide chain. CONSTITUTION:In order to elucidate specific function based on human t-PA comprising a glycoprotein containing a saccharide chain, modification of saccharide chain structure is studied and recombinant human t-PA containing alpha (1-3) bond galactose at nonreducing end of conjugate type saccharide chain is prepared with good success. This recombinant human t-PA has actions of treating plasminogen to form plasmin and has the following characteristics. (1). Specificity to synthetic substrate is as shown in the formulas. (2) Fittest pH is approximately 8-9. (3) Stable pH range is 2-10. (4) Reactivity with human natural antibody: reacting with anti-Gal antibody. Hardly being injured by existence of another oligosaccharide.
