PURPOSE:To precisely fractionating and determining LDH1 of LDH isozyme by inhibiting LDH isozyme by reacting the LDH isozyme with a protease in the presence of a protein modifier and then determining LDH1 left without being inhibited. CONSTITUTION:An isozyme is inhibited using a reaction system capable of specifically inhibiting LDH2, LDH3, LDH4 and LDH5 in a LDH isozyme group by reacting the isozyme with protease in the presence of a protein modifier such as cholic acid, guanidine (hydrochloride) or urea. LDH1 of LDH isozyme contained in a specimen such as serum or plasma is fractionated and determined by determining LDH1 left without being inhibited. Specific examples of the protease include alpha-chymotrypsin, trypsin, thrombin, elastase and endoprotease.
