Prepn. of a recombinant plasmid for the identification of allycoplasma hyopneumoniae strains and diagnosis of M. hyopneumoniae infections. The plasmid consists ECORI fragment of 2680 bp length, derived from a pUC 19 plasmid and of an ECORI fragment of 2200 bp length originating from M.hyopneumoniae. It contains an Ndel location at a distance of 1200 and 1000 bp respectively away from the ECORI location, whilst at 270 bp distance from the Ndel location. The plasma contains an ampicillin resistance glue as genetic marker. - The essence of this invention is that the DNS of the M.hyopneumoniae strain and pUC 19 plasma are digested by ECORI restrictive enzyme. The obtd. fragments are ligated and the ligation mixt. is used to transform E.coli TG-1 cells; the transformants are selected according to their ampicillin resistance and sorted by hybridising them with M.hyopneumoniae
