ANTI-SENSE TUMOR NECROSIS FACTOR MANIFESTATION VECTOR AND TUMOR NECROSIS FACTOR-SENSITIVE CELL

PURPOSE:To obtain the title cells by transformation of tumor necrosis factor- resistant tumor cells with a DNA-contg. vector capable of making said cells sensitive to tumor necrosis factor. CONSTITUTION:Firstly, using oligonucleotide synthesis process, the DNA capable of making TNF-resistant tumor cells sensitive to TNF is produced from established tumor necrosis factor (TNF) gene sequence. This DNA is bound in the direction so as to synthesize anti-sense TNFmRNA complementary to TNFmRNA, and poly A-added region is added to the downstream region, thus synthesizing a vector. Thence, tumor cells resistant to TNF is transformed with this vector to obtain the objective TNF-sensitive cells. Use of the resultant cells enables the high-sensitivity determination of TNF.
