摘要
1. The method for producing rabbit viral hemorrhagic disease (RHD) vaccine by infecting rabbits with the virulent RHD virus, getting internal organs (lungs, liver, heart, spleen, possibly kidneys and lymph nodes) from the rabbits killed at the peak of the disease symptoms, homogenizing those organs, releasing the virus by freezing and defreezing the tissue mass and centrifuging, inactivating the virus and adsorbing the antigen on the adjuvant, preferably in the form of aluminum hydroxide gel, consisting in that chloroform is gradually added while mixing in the quantity of 5-15 per cent, preferably 10 per cent by volume to the supernatant (1) obtained after the freezing, defreezing, homogenizing tissues with the addition of a buffered sodium chloride (PBS) solution and centrifuging, then it is kept at room temperature for at least one hour while mixing continuously, then the mixture is centrifuged at 3000-5000 rotations/minute and the obtained supernatant (2) is subjected to inactivation by introducing 0.1-5 per cent, preferably 0.4 per cent by volume of formalin or binary ethyleneimine, conducting the process at temperature of 20-45 degrees C, preferably 37 degrees C, then possibly adding such a quantity of PBS so that the initial tissue mass constituted 20 per cent of the final volume of the fluid after inactivation, and then the obtained antigen is adsorbed on the adjuvant used in the quantity of 5-30, preferably 15 per cent by volume in the form of aluminum hydrochloride gel.
