In the prodn. of cooked blood broth for appropriate bacteria by addn. of nicotinic acid amide adeninedinucleotide (NAD) and/or haemoglobin (Hb) as growth factors, the new feature is that blood proteins, after proteolytic degradation, are held at 60-80 deg C for 5-10 mins. then sterile: filtered conventionally. The proteolytic enzyme used is one which splits the protein into free amino acids at 25-50%. USE/ADVANTAGE - The broth is used to grow e.g. haemophilic bacteria for diagnosis (in human or veterinary medicine) and vaccine prodn. Unlike known methods, this proceudre allows large batches of broth to be produced, and eliminates the need for addn. of peptone (supplementary N source) since the blood proteins are denatured by enzyme treatment not by heating.
