New N-acetylmannosamine dehydrogenease (NAM) gene derived from a Flavobacterium species is characterised by the sequence of restriction enzyme cleavage sites S-A-S-S-Sp. (S = Sal I; A = Aat II; Sp = Sph I). Also new are (1) recombinant DNA contg. this gene in vector DNA and (2) prodn. of NAM by culturing Escherichia microorganisms contg. such recombinant DNA. USE/ADVANTAGE - NAM is a reagent used to assay sialic acid for diagnostic purposes. The new gene is efficiently expressed in E. coli without addn. of expensive inducers, so allows for simple and economic prodn. of NAM on a large scale.
