PURPOSE:To obtain the subject callus, hardly browning and capable of readily reproducing plant bodies at a high protoplast formation ratio with high reproducibility by culturing stem pieces of Ocium basilicum L. in a culture medium containing 2,4-dichlorophenoxyacetic acid for culturing plants while irradiating the stem pieces with light. CONSTITUTION:Stem pieces of Ocium basilicum L. which is a species of herbal plants are cultured in a culture medium containing 2,4-dichlorophenoxyacetic acid which is a phytohormone for culturing plants while being irradiated with light to produce a callus of the Ocium basilicum L. The resultant callus is then treated with an enzymic agent containing a cellulase formulation, pectin lyase and polygalacturonase formulation, CaCl2.2H2O and a glucide to afford protoplasts of the Ocium basilicum L. The resultant protoplasts are subsequently cultured in a culture medium containing the 2,4-dichlorophenoxyacetic acid for culturing the plants to afford plant bodies.
