METHOD FOR PREVENTING EXPRESSION OF TOLERANCE AGAINST ANALGESIC ACTION OF MORPHINE AND ITS ANALOG

PURPOSE: To obtain a pharmaceutical composition for preventing development of tolerance to analgesic action caused by continuous use of opiate, especially morphine and suppressing increase of toxicity and secondary action accompanied by increase of dosage of analgesic. CONSTITUTION: This composition for preventing development of tolerance to analgesic of morphine, scientifically related opiate, methadone and its derivative comprises GM1 monosialoganglioside, its pharmacologically permissible salt or its internal ester derivative alone or in a combination thereof as active ingredient. The composition can comprise 10-400mg active ingredient and is orally or parenterally administered at a daily dose of 0.15-6mg/kg, preferably 0.6mg/kg, preferably before administration of analgesic or simultaneously with administration of analgesic and successively with administration of analgesic over at least 8 days.
