TETRYL-BSA CONJUGATE AND PRODUCTION OF ANTI-TETRYL ANTIBODY

NEW MATERIAL:A tetryl-BSA conjugate in which a tetryl derivative having a structure expressed by the formula is bound to BSA (bovine blood serum albumin). USE:An immunogen for preparing an anti-tetryl antibody. PREPARATION:For example, tetryl having a structure expressed by the formula is dissolved in ethanol and aqueous ammonia is added thereto. Hydrogen sulfide is then dissolved in the resultant solution and reacted at 60 deg.C temperature for 24hr. The obtained reaction mixture is subjected to high-performance liquid chromatography to provide a tetryl derivative, which is subsequently dissolved in 0.5N hydrochloric acid, diluted with a phosphoric acid buffer solution (pH7.0) and reacted with a binder such as SPDP. An aqueous physiological saline solution of BSA buffered with phosphoric acid is added and reacted therewith and the prepared solution is subjected to gel filtration. Dithiothreitol is added to the active fraction and reduced to further carry out gel filtration. Thereby, the objective tetryl-BSA conjugate is obtained.
