Prodn. of low density (1.33-1.39 g/cc) heparan sulphate-proteoglycan (I) comprises first homogenising basal membrane tissue, in presence of inhibitors, at high salt concn. This step, and subsequent extraction at low salt concn., eliminates interfering proteins. The tissue residue is extracted with guanidine chloride (A) and purified by ion-exchange chromatography in presence of urea; then molecular sieve chromatography and density-gradient sepn.. Also new are highly specific sera, contg. antibodies (Ab) directed against (I) are made by immunising host animals with (I).
