NEW PROTEIN ACTING ON NERVE CELL AND GLIA CELL AND MEDICINE CONTAINING THE SAME PROTEIN

摘要

PURPOSE: To obtain the subject new protein having specific partial amino acid sequences and molecular weight and axon extension action and glia cell proliferation-suppressing action, useful as treating agent for cerebrospinal diseases such as dementia and antitumor agent for cerebral tumor. ;CONSTITUTION: A tumor lump of neurofibroma obtained from a patient of Von Recklinghausen's disease is homogenized in 20mM tris-hydrochloric acid buffer solution (pH7.5) containing a protease inhibitor and 1mM dithiothreitol and then centrifuged to separate supernatant, and the supernatant is passed through an anionic column and the absorbed ingredients are eluted with a concentration gradient of sodium chloride. Fractions having glia cell proliferation- suppressing action are collected and passed through a hydrophobic chromatography column and then purified with a hydroxyapatite column chromatography to provide the objective protein having about 50kd molecular weight, and containing partial amino acid sequences, MXR, FER, AGE, formula I, formula II, VAAAXDD, RGTPWL, GTPWLR, formula III, formula IV, ALQEAXV and formula V.;COPYRIGHT: (C)1993,JPO&Japio
