FREEZING PRESERVATION OF ADVENTITIOUS ROOT AND HAIR ROOT

PURPOSE:To preserve adventitious roots or hair roots in a high survival rate by putting specific adventitious roots or hair roots in a freezing and protecting solution comprising an aqueous solution obtained by dissolving dimethyl sulfoxide and a saccharide in a B5 basic medium, and freezing and preserving. CONSTITUTION:Adventitious roots and/or hair roots containing one or more root apexes and having =3mm length from root apex site to cut site are put in a freezing and protecting solution comprising an aqueous solution obtained by dissolving 2-10wt.% dimethyl sulfoxide and 0.1-0.5M saccharide in a B5 basic medium. After the adventitious roots or hair roots are cooled to -7 deg.C for being iced, they are precooled to at -0.5 to 2.0 deg.C/minute prefreezing and cooling rate of -30 to 40 deg.C and frozen and preserved in liquid nitrogen to freeze and preserve the adventitious roots and/or hair roots.
