KASUGAMYCIN ACETYLATION ENZYME GENE AND DNA FRAGMENT CONTAINING THE SAME

PURPOSE: To provide a new gene coding kasugamycin acetylation enzyme and useful as a selective marker for gene-introducing vector. ;CONSTITUTION: The objective gene coding a kasugamycin acetylation enzyme is effective to impart a host cell with a capability to produce a kasugamycin acetylation enzyme to inactivate kasugamycin and make a host cell to be resistant to kasugamycin. The gene is e.g. the DNA fragment having a molecular weight of about 14 kb and originated from the DNA of actinomycete and is present in the filled part of the cleavage map. It can be prepared by separating a DNA fragment having a molecular weight of about 14kb and containing a gene to produce an enzyme to acetylate and inactivate kasugamycin from various DNA fragments obtained by cleaving total DNA in a cell of Streptomyces kasugaensis MB273 strain with restriction enzyme PstI, integrating the DNA fragment into a plasmid and cloning the product.;COPYRIGHT: (C)1993,JPO&Japio
