described is a process for the production of penicillin acylase in high yield. the bacillus subtilis - plasmid used in this chromosomales dna fragment from bacillus megaterium is fitted. this is a suitable gene to increase the rate of penicillin acylase.the recombinant plasmid is introduced into bacillus megaterium and thus transformed cells to produce penicillin acylase used. moreover, a plasmid, a method of manufacturing the same and a host plasmid combination provided in the above procedures can be used.
