Treatment of allergic rhinitis using pAF antagonists - e.g. thieno-, cyclopenta-(4,5)-thieno- or 2,3,4,5-tetra:hydro-8H-pyrido-(4,5)thieno-(3,2-f) (1,2,4) triazolo-(4,3-a) (1,4) diazepine

PAF antagonists of hetrazepinoid structure (I) are used in the prophylaxis and therapy of allergic rhinitis and allergic conjunctruitis. (I) is pref. a deriv. of a thieno (3,2-f) (1,2,4) triazolo-(4,3-a) (1,4) diazepin, a cyclopenta (4,5) thieno (3,2-f) (1,2,4) triazolo (4,3-a) (1,4) diazepin, or a 2,3,4,5-tetrahydro-8H-pyriodo -(4,5) thieno (3,2-f) (1,2,4) - triazolo (4,3-a) (1,4) diazepin, esp. WEB 2086, WEB 2170, Y20411, Y 24180, RO 244736, E6123, BN50730, BN50739 or BN50726. USE/ADVANTAGE - (I) can be used to treat allergic reactions of the nasal mucons membrances and of the conjunchiva, replacing the use of e.g antihistanines and resulting in fewer side effects. (I) can be used to treat perenvial and seasonal rhinits and can be used orally as a prophylactic, esp if taken 10-14 days before the onset of the pollen season. Suitable oral doses are 10-100 mg taken 3 times daily. (I) can also be used as a nasal spray, eg as a 0.3% aq.soln. providing approx. 0.21 mg per dose. (I) can also be used parenterally, as suppositories or as eye drops.
