PURPOSE:To obtain the title enzyme useful for a substrate to assay-N-accethyl glucosaminidase in urine by making a culture of a specific strain classified as Pseudomonas sp. CONSTITUTION:A culture medium is inoculated with a strain classified as Pseudomonas sp., capable of producing N-acethyl hexosamine dehydrogenase [e.g., Pseudomonas sp. No. 5353 (FERM BP-2057)] to make a culture either at 20-35 deg.C for 20-30hr under shaking or deep stirring to deprive N-acethyl glucosamine or N-acethyl galactosamine of hydrogen into N-acethyl glucosaminolactone or N-acethyl galactosaminolactone, respectively, and, at the same time, to obtain the objective N-acethyl hexosamine dehydrogenase which reduces a coenzyme NAD into NADH.
