HOMOGENEOUS ENZYME IMMUNOASSAY, ENZYME-LABELED ANTIBODY AND DRY IMMUNOASSAYABLE ELEMENT

PURPOSE: To provide a homogenous enzymatic immunoassay which is simple and provides high sensitivity be employing a dry immunoassayable element containing an enzyme-labeled substance consisting of the bonded both of an enzyme with two monoclonal antibodies or more which recognize antigenic determinating groups for different antigens. ;CONSTITUTION: Ligands contained in a sample and having antigen determining groups, i.e., antigens are employed as subjects for measurement, and two monoclonal antibodies or more which recognize antigen determinating groups for different antigens for measurement are employed. Enzymes which form bonded body with the antibodies are selected on the basis of subsequent enzyme reaction, and the functional groups of two material are used in bonding the enzymes and the antibodies, and a support 10, a detecting layer (or reagent layer) 12 and an immunoreaction layer 14 are stacked one stop another to form a dry immunoassayable element. When the sample is attached to the reaction layer 14, the sample (antigen) undergoes an antigen-antibody bonding reaction with the enzyme-labeled antibodies and forms a matrix structure, thus suppressing the enzyme activity to the matrix inclusion. Therefore, the amount of antigens in the sample can readily and accurately be measured by measuring the amount of enzymatic reaction products at the reagent layer 12.;COPYRIGHT: (C)1994,JPO&Japio
