Chemiluminescent tissue or cell analysis - using peroxidase labelling, with signal detection by photon microscope and camera for high sensitivity

Analysis of tissue or cells comprises: (a) contacting a previously prepd. cytological or histological sample with a chemiluminescent reactant, consisting of a luminogenic substrate for a peroxidase enzyme in combination with an amplifying agent; (b) immediately carrying out the analysis; and (c) collecting the luminescent signal with a photon microscope equipped with an immersion lens, the output from which is supplied to an ICCD camera linked to a control unit and an image processor provided with a photon counter, the analogue image being visualised on a video monitor or another visualisation on a video monitor or another visualisation treatment or recording device. The samples are pref. obtd using a nucleic acid probe for in situ hybridisation using a specific primary antibody. The probe is traced by an immunoassay using a peroxidase as enzymatic label. The chemiluminescent reactant is luminol, contg. a luminogenic substrate for peroxidase in combination with p-iodophenol as amplifying agent. The samples are kept in aq. media.
