PURPOSE:To produce a large number of cloned seedlings of American carrot in high efficiency, by inducing callus from a part of the living tissues, forming shoots from the callus and propagating them into multiple shoots. CONSTITUTION:A part of living tissues such as roots, stems, leaves, or flower buds are cut off, sterilized in a sodium hypochlorite solution containing Tween 80 and tissue-cultured in a callus inducing medium containing auxin such as 2,4-D or cytokinin such as MURASHIGE-SCOUGUE medium to form callus. Then, the callus is cultured in an adventitious embryo-inducing medium to induce the embryos. The matured embryos among the induced embryos are cultured in a redifferentiation medium containing cytokinin or gibberellin under irradiation with light to form bud shoots in about 4 weeks. The bud shoots are cultured in a medium containing BAP-GA or the like under irradiation with light to form multiple shoots. Thus, the mass production of seedlings of American carrot becomes possible in high efficiency.
