DETERMINATION OF NUCLEASE ACTIVITY AND REAGENT FOR DETERMINATION OF NUCLEASE ACTIVITY

PURPOSE: To provide an easy, quick, sensitive and safe method for the determination of nuclease activity and a reagent to be used for the determination. ;CONSTITUTION: A dilution series of RNase with BSA(bovine serum albumin) is used as a specimen. The specimen is mixed with a buffer solution (Tris-HCI and EDTA.2Na), preincubated at 25°C for 6min and incorporated with a substrate liquid containing a fluorescent pigment thioflavin T and RNA to start the reaction. The fluorescent intensity decrement (ΔI) is determined with a spectrofluorophotometer for 20min accurately from 1min to 21min after the start of the reaction.;COPYRIGHT: (C)1994,JPO
