METHOD FOR TISSUE CULTURE OF ROOT OF WOODY PLANT

PURPOSE: To facilitate elongation of roots in a radiated state by culturing seedling materials in a callus inducing medium in a short period of time, taking out the seedling materials before they are completely callused, removing stems and leaves, transplanting the seedling materials in a medium for elongation, elongating the roots and efficiently carrying out tissue culture of the roots. ;CONSTITUTION: For example, a seed of mulberry is germinated in a medium for germination and about four to eight seedling materials are adjoined to a callus inducing medium. After 10 or 20 days, the seedling materials are taken out before they are completely callused. The seedling materials from which stems 3 and leaves 4 are removed are transplanted in a medium for elongation which is prepared by blending B-5 medium as a basal medium with 2.5g/l gellan gum as a coagulant and adjusting to pH 6.00 and roots 2 are elongated to carry out tissue culture of the roots 2.;COPYRIGHT: (C)1995,JPO
