Synthesis of binding sites between two proteins - by determn. of hydropathic values, e.g. synthesis of HIV-1 Gp120 and CD4 receptor complementary sequences
Synthesis of peptides (A' and B') identified as the active binding sites between two proteins (A and B) pref. both of natural origin, is new, wherein (a) the amino acid (AA) sequence of (A) and (B) is determined, (b) the hydropathic value (HPV) for each element of the sequence of (A) and (B) is determined by means of a predetermined scale of HPVs (given in the specification. (C) the HPVs are summed according to the formula, (I)-(IV) r = integer defined as the run length (r is less than), S = (r-1)/2 per each residue of (A) and (B), (d) the complementary HP score for a given search length L is calculated according to the formula (V), L = lower than the total number of AAs comprised in the proteins, (e) at least a pair of sequences of length L for the interacting couple, exhibiting the lowest score, are selected and (f) the formed sequences are synthesised. Also claimed is an affinity column for purifying proteins and peptides binded on suitable solid support.
