Method for making a synthetic oligonucleotide which binds to a target sequence in duplex DNA forming, by binding to the major groove, a colinear triplex, said method comprising the steps of: identifying within a genomic DNA double helix of a nucleotide target sequence of about more than 20 nucleotides, containing either at least 65% purine bases about or at least 65% pyrimidine approximately bases; and synthesizing said complementary to said identified target sequence synthetic oligonucleotides, wherein said synthetic oligonucleotides one G when the complementary location in the DNA double helix exhibits a GC base pair, and having a T when the complementary location in the DNA double helix exhibits an AT base pair.
