MEROCYANINE-BASED LABELING COLORING MATTER, ITS PRECURSOR COMPOUND, SYNTHESIS OF MEROCYAMINE-BASED LABELING COLORING MATTER AND MEROCYANINE-LABELED PROTEIN

PURPOSE: To obtain the subject labeling coloring matter, having isothiocyanyl bindable to primary and secondary amino groups in an aqueous solution, reactive with amino groups in a protein and useful for preparing a merocyanine label. CONSTITUTION: This merocyanine-based labeling coloring matter comprises an isothiocyanate derivative of formula I (X is a halogen). The isothiocyanate derivative can be obtained by stirring an amino derivative of formula II which is its precursor with thiocarbonyldiimidazole in a mixed polar organic solvent and adding the resultant reactional product into a nonpolar solvent. The derivative of formula II can be obtained by mixing and heating, e.g. bromopropylphthalimide with trimethylindolenine, reacting the resultant trimethylindolenium salt with dimethylaminobenzaldehyde and providing the derivative of formula II from the prepared phthalimide derivative and hydrazine monohydrate. Furthermore, the coloring matter is capable of labeling a protein such as an antibody, albumin in blood, etc., having about 150000 molecular weight and carrying out the accurate determination.
