SYNTHESIS OF ACTIVATED PROTEIN IN CELL-FREE PROTEIN SYNTHESIS SYSTEM

PURPOSE: To obtain the objective protein holding the right structure and having increased specific activity by removing a reducing agent from a synthetic reaction liquid of a cell-free protein synthesis system and keeping the system to an oxidizing condition. ;CONSTITUTION: A reducing agent is removed from a synthetic reaction liquid of a cell-free protein synthesis system by the salting-out treatment and subsequent dialyzing treatment and the system is maintained in an oxidizing state. The dialyzing treatment is carried out e.g. by using a cellophane tube, pouring a cell-free protein synthesis reaction liquid into the tube and dialyzing the liquid using a buffer solution of pH1-10 as the outer dialysis solution.;COPYRIGHT: (C)1997,JPO
