Rabbit liver NADPH cytochrome P450 reductase gene

摘要

PURPOSE:To separate a gene coding a rabbit liver NADPH-cytochrome P-450 reductase reactive to cytochrome P-450 playing important role in metabolism of a foreign fat-soluble compound such as drug in living body. CONSTITUTION:mRNA is prepared from liver microsome of a rabbit administered with phenobarbital. A fraction containing an mRNA of rabbit liver NADPH-cytochrome P-450 reductase is prepared from the above mRNA by using a part of a rat liver NADPH-cytochrome P-450 reductase gene having known structure as a probe. A cDNA is prepared therefrom by Okayama-Berg method and is introduced into E.coli. A desired cDNA is selected by the use of the above probe and the separation can be ascertained by determining the base sequence.
