The method provides for the use of Cupressus arizonica pollen harvested by gravity under optimum climatic conditions, selected on the basis of macroscopic and microscopic criteria and having a degree of purity of not less than 99.5%. This pollen is then preserved from deterioration by freezing at -30 degree C. The method also provides for the preparation of an allergenic extract by double extraction using two different concentrations of NH4HCO3 and subsequent concentration of the extract by precipitation with (NH4)2SO4. The allergenic extract so obtained is then used to prepare an ELISA test which is capable of detecting the presence of Cupressaceae-antigen counter-IgE in at least 75% of individuals affected by such pollen. The test is also highly specific.
