FIELD: biotechnology, molecular biology, genetic engineering. SUBSTANCE: invention relates to the chemical synthesis of oligonucleotide probes on the solid polymeric carrier. Method involves splacing the required nucleotide number, blocking hydrophosphoryl internucleotide bonds followed by addition of the next nucleotide to the synthesized fragment and aliphatic amine (substance-spacer of the required length) to the formed hydrophosphoryl bond. Then biotin label is linked to free functional group of substance-spacer and nucleotide splacing is continued up to the next label addition. During isolation and purification of probe hydrophosphoryl internucleotide bonds were deblocked. Addition of biotin labels to hydrophoisphoryl internucleotide bonds by substance-spacer use does not destroy nitrogen bases structure of the synthesized oligonucleotide that provides the complete identity of the synthesized probe to complementary site of examined nucleic acid.
