PURPOSE: To classify and count leucocytes in a sample to a minimum of eight by including a process for measuring scattered light intensity of leucocytes and with influence of erythrocytes in a hematological sample eliminated and a process for specifically dyeing leucocytes and erythroblasts. ;CONSTITUTION: Aqueous solution of low osmotic pressure comprising a buffer material for maintaining pH of the solution in an acidic region and a hematological sample are mixed to fraction erythrocytes in the sample to damage erythroblast films. Then osmatic pressure compensating agent for maintaining forms of leucocytes and solution comprising buffer agent which neutralizes with acid to make the solution at applicable pH for dyeing are added to the solution. Then the leucocytes contained in the sample are dyed by four pigments including astrazon yellow 3G for dyeing halophilic radical cells and young granule cells, neutral red being dyeing pigment for acidophilic cells, a pigment for dyeing a nucleus or cytoplasma or both of a leucocyte and a fluorescent pigment for dyeing only a nucleus which was damaged. Thus two groups comprising young granule cells, erythroblasts, halophilic cells, acidiphilic cells, lymphocytes, monocytes and neutrophils can be classified and counted.;COPYRIGHT: (C)1993,JPO&Japio
