PURPOSE:To measure PTHrP simply in high sensitivity and to make it possible to utilize this method for differential diagnosis of hypercalcemia (HHM) by making an antigen, which specifically recognizes the N-end part and the intermediate part of PTHrP, act on a specimen, and detecting the generated antibody-antigen-antibody reaction. CONSTITUTION:PTHrP is decomposed into an N-end part, an intermediate part and a C end in a bloodstream. Therefore, an antigen which recognizes the N-end part and the intermediate part is made to act. The fragments of the N-end part and the intermediate part used as the antigens for preparing the N-end antibody and the intermediate-part antibody have the amino-acid chain lengths of about 20-35 pieces. The N-end antibody and the intermediate-part antibody are used, and either of these antibodies is labeled and sequentially made to react. Thus, the antibody-label complex of N-end antibody, antigen and intermediate antibody is generated. The antibody which is not reacted is removed from the complex, and the radiation activity, enzyme activity and fluorescence activity of the composite material in the complex are measured. Thus the amount of the labels is measured. Finally, the amount of PTHrP is obtained from the measured amount of the labels, and this value is made to be the measured value.
