PURPOSE:To regenerate the protoplast of biffidobacterium in a high yield by smearing the biffidobacterium protoplast on a specific composition medium mixed an osmotic stabilizer and subsequently culturing the protoplast under an anaerobic condition. CONSTITUTION:MNM-G medium having a composition 2.5g of K2HPO4, 10g of CH3COONa, 10g of tryptone, 5.0g of a yeast extract, 0.4g of cysteine.HCl.H2 O, 5.0g of glucose, 15g of a bact agar, 5mM of CaCl2.2H2O, 5mM of MgCl2.6H2 O and a pH of 6.5 is mixed with an osmotic stabilizer raffinose in a concentra tion of 0.2-0.3M. A biffidobacterium.pseudolongum or biffidobacterium.longum protoplast suspension diluted with a hypertonic buffer solution is smeared on the prepared medium and cultured under an anaerobic condition.
